Samuel was the eldest son of Samuel and Susannah Wesley and older brother of John and Charles. He was born in Spitalfields, London in 1690/1.
He was educated at Westminster School before entering Christ Church, Oxford in 1711 where he received a BA degree in 1715 and MA in 1718. After his ordination he became teacher at Westminster School. During this time his younger brother Charles attended the school as a pupil.
In 1733 he was appointed master of Tiverton Grammar School (Blundells) in Devon where he died on 6 November 1739. He is buried in Tiverton churchyard.
Samuel married Ursula Berry (died 1742), daugher of Rev John Berry, vicar of Walton, Norfolk. They were survived by a daughter and a son, Samuel.
